The enzymatic conversion of 5-formyluracil to uracil 5-carboxylic acid.

چکیده
Fink and Fink (1) have shown that Neurospora crassa mycelia convert thymidine to the uracil and cytdsine of RNA without fragmentation of the pyrimidine ring. A cell-free system from Neurospora catalyzes several reactions which appear (l-3) to be involved in this pathway effecting the removal of the methyl group of thymidine. These reactions are the conversions of thymidine to thymine ribonucleoside (4), thymine to 5-hydroxymethyluracil (5), and the latter compound to 5-formyluracil (6j.l In studies with more active preparations of this enzyme system (4), the 5-formyluracil, which was generated, appeared occasionally to be converted to a very small amount of a new compound. This report documents the enzymatic conversion of 5.formyluracil to uracil5-carboxylic acid.

Metabolism of pyrimidine deoxyribonucleosides in Neurospora crassa.
The experiments in this report involve the following series of reactions which were previously demonstrated with purified enzyme preparations from Neurospora crassa: thymidine a yields thymine ribonucleoside b yields thymine c yields 5-hydroxymethyluracil d yields 5-formyluracil e yields uracil-5-carboxylic acid f yields uracil. The evidence for some of the reactions occurring in vivo has been ...
متن کاملMutants affecting thymidine metabolism in Neurospora crassa.
When (14)C-thymidine labeled only in the ring is administered to Neurospora crassa, the majority of the recovered label is found in the ribonucleic acid (RNA). Three mutants were isolated in which different steps are blocked in the pathway that converts the pyrimidine ring of thymidine to an RNA precursor. Evidence from genetic, nutritional, and accumulation studies with the three mutants shows...
متن کاملOpposite-base dependent excision of 5-formyluracil from DNA by hSMUG1.
PURPOSE The aim of this study was to determine the excision efficiency of hSMUG1 (human single-strand-selective monofunctional uracil-DNA glycosylase) for 5-formyluracil (fU), a major thymine lesion formed by ionizing radiation, opposite all normal bases in DNA, to possibly explain mutation induction by fU in the DNA of mammalian cells. MATERIALS AND METHODS An enzymatically [(32)P]labelled f...
متن کاملMutational analysis of the damage-recognition and catalytic mechanism of human SMUG1 DNA glycosylase.
Single-strand selective monofunctional uracil-DNA glycosylase (SMUG1), previously thought to be a backup enzyme for uracil-DNA glycosylase, has recently been shown to excise 5-hydroxyuracil (hoU), 5-hydroxymethyluracil (hmU) and 5-formyluracil (fU) bearing an oxidized group at ring C5 as well as an uracil. In the present study, we used site-directed mutagenesis to construct a series of mutants ...
متن کاملIdentification of high excision capacity for 5-hydroxymethyluracil mispaired with guanine in DNA of Escherichia coli MutM, Nei and Nth DNA glycosylases.
The oxidation and deamination of 5-methylcytosine (5mC) in DNA generates a base-pair between 5-hydroxymethyluracil (5hmU) and guanine. 5hmU normally forms a base-pair with adenine. Therefore, the conversion of 5mC to 5hmU is a potential pathway for the generation of 5mC to T transitions. Mammalian cells have high levels of activity of 5hmU-DNA glycosylase, which excises 5hmU from DNA. However, ...
